Adamantane

Adamantane is a saturated hydrocarbon, C₁₀H₁₆, whose rigid cage of ten carbon atoms reproduces a fragment of the diamond lattice.

Cyclohexane: A saturated six-carbon ring whose conformations illustrate the bond geometry found in many hydrocarbons. Its chair conformation provides a familiar reference for the tetrahedral bonds in adamantane.
IUPAC nomenclature: A standardized system for naming chemical compounds and describing their structures. Adamantane’s name is retained as the parent for many substituted cage derivatives.
Souto petroleum: A petroleum source in Czechoslovakia from which adamantane was first isolated. The first natural sample of adamantane came from this petroleum.
Amantadine: A medication containing an adamantane group, used for Parkinson’s disease and historically for influenza A. It is a prominent drug derived from the adamantane scaffold.
Cubane: A hydrocarbon, C₈H₈, whose eight carbon atoms occupy the corners of a cube. Cubane is a more highly strained cage, unlike adamantane’s near-ideal tetrahedral framework.
Tetrahedral molecular geometry: A molecular shape in which a central atom’s four bonds point toward the corners of a tetrahedron. Each carbon in adamantane has four tetrahedrally arranged bonds.
Conformational analysis: The study of molecular shapes accessible through rotations around single bonds. Adamantane’s fused cage restricts the conformational freedom typical of flexible hydrocarbons.
Stanislav Landa: A Czech chemist who isolated adamantane from petroleum in the 1930s. Landa identified the compound that became known as adamantane.
Memantine: A medication used to treat moderate to severe Alzheimer’s disease. Its adamantane-based structure helps illustrate the cage’s role in drug design.
Twistane: A polycyclic hydrocarbon with a twisted carbon framework and formula C₁₀H₁₆. It shares adamantane’s formula but has a different cage connectivity and shape.
