Adenosine receptor antagonist

An antagonist that binds to adenosine receptors and reduces or prevents their activation by adenosine. These substances can alter signaling involved in sleep, vascular tone, inflammation, and cardiac activity.

Adenosine receptor: A cell-surface receptor activated by adenosine, with subtypes A1, A2A, A2B, and A3. Antagonists work by preventing adenosine from activating these receptors.
Caffeine: A methylxanthine alkaloid that promotes wakefulness chiefly by antagonizing adenosine receptors at ordinary doses. It is the best-known adenosine antagonist and a widely consumed stimulant.
Adenosine: A purine nucleoside formed in cells and extracellular spaces that acts as a signaling molecule. It is the endogenous ligand whose receptor effects antagonists oppose.

Theophylline: A methylxanthine drug used mainly as a bronchodilator, with adenosine-receptor antagonism among its actions. Its adenosine blockade contributes to both therapeutic effects and toxicity.
Adenosine triphosphate: A nucleotide that stores and transfers chemical energy and can be released outside cells as a precursor to adenosine. Extracellular ATP breakdown helps generate the adenosine that activates target receptors.
Adenosine reuptake inhibitor: A substance that reduces cellular uptake of adenosine, increasing its extracellular availability. It strengthens endogenous adenosine signaling rather than blocking its receptors.
Inverse agonist: A ligand that reduces a receptor’s constitutive activity by favoring its inactive state. Some receptor blockers can suppress baseline signaling, unlike neutral antagonists.
Istradefylline: A selective A2A adenosine receptor antagonist approved as an adjunct treatment for Parkinson’s disease. It applies subtype-selective antagonism to reduce off episodes in Parkinson’s disease.
