Barbituric acid

Barbituric acid is a heterocyclic organic compound whose derivatives include barbiturate drugs. The parent compound itself has little pharmacological activity.

Pyrimidine: A six-membered aromatic heterocycle containing two nitrogen atoms. Barbituric acid has a pyrimidine-derived ring framework.
Biginelli reaction: A multicomponent reaction that forms dihydropyrimidinones from an aldehyde, a β-ketoester, and urea or thiourea. It is a distinct route to related pyrimidine-containing products, not the classical barbituric-acid synthesis.
Barbiturate: A compound derived from barbituric acid, many of which depress activity in the central nervous system. Barbiturates are the drug family built around substitutions on this parent scaffold.
Pyrimidinone: A pyrimidine derivative containing a carbonyl group within its ring. It is a related carbonyl heterocycle but lacks barbituric acid's two nitrogen-adjacent carbonyl groups.
Urea: A small organic compound with a carbonyl group bonded to two amino groups. Urea supplies the nitrogen-containing component in the classical synthesis of barbituric acid.
Claisen condensation: A carbon–carbon bond-forming reaction between esters, promoted by a base. The classical preparation uses a condensation of diethyl malonate with urea.
Phenobarbital: A long-acting barbiturate used as an antiseizure medicine and, less often, as a sedative. Its substitutions illustrate how modifying the parent structure produces pharmacologically active drugs.
Hydantoin: A five-membered nitrogen-containing heterocycle with two carbonyl groups. Hydantoins can also yield anticonvulsant drugs, but have a smaller ring and different scaffold.
Malonic acid: A dicarboxylic acid with two carboxyl groups attached to one carbon atom. Its carbon skeleton provides the carbon framework in the classical ring-forming reaction.
Diethyl malonate: The diethyl ester of malonic acid, commonly used in carbon–carbon bond-forming reactions. It reacts with urea to form the barbituric-acid ring.
