Biological half-life

Biological half-life is the time required for an organism or tissue to eliminate half of a substance’s amount. It depends on the substance, biological system, and conditions of measurement.

Exponential decay: A process in which a quantity decreases at a rate proportional to its current value. A constant biological half-life produces exponential decline when elimination follows first-order kinetics.
Drug dosing: The selection of drug amounts and timing to achieve a desired therapeutic exposure. Half-life helps set dosing intervals and predict how quickly repeated doses accumulate.
Radioactive half-life: The time required for half the unstable nuclei in a sample to decay. Radioactive half-life is a nuclear property, unlike biological half-life, which depends on organism-level removal.
Caffeine metabolism: The enzymatic breakdown and elimination of caffeine, chiefly involving liver enzymes. Caffeine’s biological half-life varies with factors that alter its hepatic metabolism.
First-order kinetics: Kinetics in which the rate of a process is proportional to the concentration of a reactant. This concentration-dependent elimination yields a constant half-life.
Steady state (pharmacology): A condition in which the rate of drug administration equals its rate of elimination over time. Repeated dosing commonly approaches steady state after several biological half-lives.
Effective half-life: The time for a radioactive substance in an organism to decrease by half through both physical decay and biological elimination. It combines biological elimination with radioactive decay rather than measuring elimination alone.
Lithium pharmacokinetics: The absorption, distribution, and renal elimination of lithium used as a medication. Lithium illustrates how kidney function strongly influences a drug’s biological half-life.
Pharmacokinetics: The study of how organisms absorb, distribute, metabolize, and excrete substances. Biological half-life summarizes one part of a substance’s movement through the body.
Loading dose: An initial drug dose used to reach a target concentration more rapidly than maintenance dosing alone. A long half-life can make a loading dose useful when rapid therapeutic levels are needed.
