Clomifene

Clomifene is a selective estrogen receptor modulator used mainly to induce ovulation in some forms of infertility. It stimulates reproductive hormone release by blocking estrogen feedback in the hypothalamus.

Hypothalamic–pituitary–gonadal axis: The hormone-signaling system linking the hypothalamus, pituitary gland, and gonads. Clomifene acts by altering estrogen feedback within this reproductive control system.
Anovulation: The absence of ovulation during menstrual cycles. Clomifene is used when absent or infrequent ovulation contributes to infertility.
Tamoxifen: A selective estrogen receptor modulator used mainly in hormone-receptor-positive breast cancer. It shares clomifene’s drug class but has a different principal clinical use.
Ovarian hyperstimulation syndrome: A complication of fertility treatment in which enlarged ovaries and fluid shifts cause symptoms ranging from mild to severe. It is a fertility-drug risk to consider, though less common with clomifene than injectable gonadotropins.
Gonadotropin-releasing hormone: A hypothalamic hormone that prompts the pituitary gland to release LH and FSH. Reduced estrogen feedback can increase its pulsatile secretion during clomifene treatment.
Polycystic ovary syndrome: A hormonal and metabolic condition often associated with irregular ovulation and androgen excess. It is a common cause of anovulatory infertility for which clomifene may be considered.
Toremifene: A selective estrogen receptor modulator used in some cases of hormone-receptor-positive breast cancer. It provides another comparison for how members of clomifene’s drug class are used.
Hot flashes: Sudden episodes of warmth, often accompanied by flushing and sweating. They are a common temporary side effect of clomifene’s anti-estrogenic activity.
Follicle-stimulating hormone: A pituitary hormone that supports ovarian follicle development and sperm production. Higher FSH helps recruit and mature ovarian follicles during treatment.
Infertility: A reproductive condition in which pregnancy does not occur after a period of regular unprotected intercourse. Clomifene addresses selected ovulatory causes, not every form of infertility.
