Daunorubicin

Daunorubicin is an anthracycline chemotherapy drug used mainly to treat acute leukemias. It damages DNA by intercalating into it and inhibiting topoisomerase II.

DNA intercalation: The insertion of a planar molecule between adjacent base pairs in a DNA helix. Daunorubicin intercalates into DNA, disrupting replication and transcription.
Acute myeloid leukemia: A rapidly progressing cancer in which abnormal myeloid cells accumulate in bone marrow and blood. Daunorubicin is a standard component of induction chemotherapy for many patients with this leukemia.
Cardiotoxicity: Heart damage caused by exposure to a drug, toxin, or other harmful factor. Cumulative daunorubicin exposure can cause irreversible cardiac dysfunction.
Doxorubicin: An anthracycline chemotherapy drug used against a range of solid tumors and blood cancers. It shares daunorubicin’s core drug class but is used across a different set of cancer regimens.
Topoisomerase II: An enzyme that changes DNA topology by cutting both strands, passing another DNA segment through the break, and resealing it. Daunorubicin traps this enzyme on DNA, creating breaks that cells struggle to repair.
Acute lymphoblastic leukemia: A rapidly progressing cancer of immature lymphoid cells in bone marrow and blood. Daunorubicin is used in multi-drug treatment regimens, particularly for high-risk disease.
Anthracycline-induced cardiomyopathy: A weakening of the heart muscle associated with anthracycline chemotherapy. This is the characteristic long-term cardiac complication that limits daunorubicin dosing.
Idarubicin: An anthracycline chemotherapy drug used in acute leukemia treatment. It can replace daunorubicin in acute myeloid leukemia induction regimens.
Reactive oxygen species: Chemically reactive oxygen-containing molecules that can damage cellular components. Daunorubicin can generate these molecules, contributing to tissue injury, especially in the heart.
Cytarabine: An antimetabolite chemotherapy drug that interferes with DNA synthesis. It is commonly combined with daunorubicin during induction treatment for acute myeloid leukemia.
