Famotidine

Famotidine is a histamine H₂-receptor antagonist that reduces stomach acid secretion. It is used to treat conditions including peptic ulcers and gastroesophageal reflux disease.

Histamine H₂ receptor: A G protein-coupled receptor through which histamine stimulates gastric acid secretion. Famotidine reduces acid by blocking this receptor on stomach parietal cells.
Gastroesophageal reflux disease: A condition in which stomach contents repeatedly flow into the esophagus, causing troublesome symptoms or complications. Famotidine can reduce the acidity of reflux and relieve some associated symptoms.
Proton-pump inhibitor: A medicine that suppresses gastric acid by inhibiting the stomach’s H⁺/K⁺-ATPase proton pump. This drug class generally produces stronger, longer-lasting acid suppression than famotidine.
Pharmacokinetics: The study of how a drug is absorbed, distributed, metabolized, and eliminated by the body. Famotidine’s absorption and kidney clearance shape its dosing and duration.
James Black: A Scottish pharmacologist whose receptor-based drug research led to major therapeutic medicines. His work on histamine antagonists established the path toward H₂-blocking drugs.
Parietal cell: A stomach epithelial cell that secretes hydrochloric acid into the gastric lumen. These cells are the principal target cells for famotidine’s acid-reducing action.
Peptic ulcer disease: A disorder involving open sores in the stomach or duodenal lining. Reducing acid can support healing of ulcers alongside treatment of their underlying cause.
Cimetidine: A histamine H₂-receptor antagonist used to reduce gastric acid secretion. It belongs to the same drug class but has more clinically significant drug interactions.
Renal impairment: Reduced kidney function that can limit the elimination of substances from the body. Famotidine is cleared largely by the kidneys, so impaired function can require dose adjustment.
Burimamide: An early experimental histamine H₂-receptor antagonist developed during the search for acid-suppressing drugs. It was a predecessor in the series of compounds that led to famotidine.
