Flunitrazepam

Flunitrazepam is a potent benzodiazepine sedative and hypnotic used medically for severe insomnia and, in some countries, anesthesia. It can cause dependence and impair memory.

GABA receptor: A receptor family that responds to gamma-aminobutyric acid, the main inhibitory neurotransmitter in the brain. Flunitrazepam acts at GABA-A receptors, amplifying the brain’s inhibitory signaling.
Insomnia: A persistent difficulty falling asleep, staying asleep, or obtaining restorative sleep despite adequate opportunity. Flunitrazepam has been prescribed for severe insomnia in some countries.
Benzodiazepine dependence: Physical or psychological reliance on benzodiazepines, sometimes accompanied by tolerance and withdrawal symptoms. Repeated flunitrazepam use can lead to dependence, especially with prolonged or high-dose use.
Diazepam: A long-acting benzodiazepine used for anxiety, muscle spasms, seizures, and alcohol withdrawal. It shares flunitrazepam’s receptor mechanism but has different clinical uses and duration.
GABA-A receptor: A ligand-gated chloride channel that mediates fast inhibitory signaling in the central nervous system. This receptor is the direct target through which flunitrazepam produces sedation.
Hypnotic: A substance used to induce or maintain sleep. Flunitrazepam’s principal therapeutic role is as a potent sleep-inducing drug.
Benzodiazepine withdrawal syndrome: A set of symptoms following abrupt reduction or cessation after sustained benzodiazepine use. Stopping flunitrazepam suddenly after regular use can provoke withdrawal, including seizures in severe cases.
Nitrazepam: A benzodiazepine hypnotic prescribed for short-term treatment of severe insomnia in some countries. It is a closely related sleep medication with overlapping effects and a different chemical structure.
Benzodiazepine site: An allosteric binding site on GABA-A receptors where benzodiazepines modulate channel activity. Flunitrazepam binds here and increases GABA’s effects without directly opening the channel.
Premedication: Medication given before a procedure to reduce anxiety, discomfort, or awareness. Flunitrazepam has been used before procedures because it causes sedation and amnesia.
