Glimepiride

Glimepiride is an oral sulfonylurea medication used with diet and exercise to lower blood glucose in people with type 2 diabetes. It stimulates insulin release from pancreatic beta cells.

Sulfonylurea receptor 1: A regulatory subunit of ATP-sensitive potassium channels in pancreatic beta cells that binds sulfonylurea drugs. Glimepiride binds this channel subunit, helping trigger insulin release.
Type 2 diabetes: A chronic metabolic disease characterized by high blood glucose resulting from insulin resistance and progressive beta-cell dysfunction. Glimepiride is prescribed as an adjunct to diet and exercise for this form of diabetes.
Glipizide: A short-acting sulfonylurea used to lower blood glucose in type 2 diabetes. Its shorter action can make meal timing and hypoglycemia patterns differ from glimepiride.
Weight gain: An increase in body mass that can result from changes in energy balance, fluid status, or medication effects. Sulfonylurea-stimulated insulin release can promote weight gain during glimepiride treatment.
ATP-sensitive potassium channel: A membrane channel that links cellular energy levels to electrical activity by controlling potassium flow. Glimepiride closes beta-cell channels of this type, initiating the sequence that releases insulin.
Metformin: An oral glucose-lowering medication that primarily reduces hepatic glucose production and improves insulin sensitivity. It is commonly preferred as initial drug therapy, whereas glimepiride increases insulin secretion.
Glyburide: A sulfonylurea medication that stimulates insulin release to treat type 2 diabetes. Glyburide is associated with greater prolonged hypoglycemia risk than glimepiride, particularly in older adults.
Cardiovascular disease: A group of disorders affecting the heart and blood vessels, including coronary artery disease and stroke. Cardiovascular risk influences diabetes drug selection and interpretation of glimepiride outcomes.
Insulin secretion: The release of insulin from pancreatic beta cells in response to nutrients and other signals. This is the immediate physiological process glimepiride amplifies.
Glycated hemoglobin: Hemoglobin with glucose attached, measured as a marker of average blood glucose over roughly two to three months. Clinicians use this measure to assess longer-term response to glimepiride.
