Gram-positive bacteria

Gram-positive bacteria retain the crystal violet–iodine complex during Gram staining, appearing purple. Their envelopes typically have thick peptidoglycan walls and lack the outer membrane characteristic of Gram-negative bacteria.

Peptidoglycan: A mesh-like polymer of sugars and peptides that provides bacterial cell walls with strength and shape. Its thick layer usually traps the crystal violet–iodine complex in these bacteria.
Iodine: A chemical element used in Gram staining to form a complex with crystal violet. The crystal violet–iodine complex is larger and less readily removed from thick walls.
Bacterial culture: The growth of bacteria in controlled conditions for study, detection, or identification. Cultured isolates can be examined by Gram stain as an early identification step.
Beta-lactam antibiotics: Antibiotics that inhibit bacterial cell-wall synthesis by targeting penicillin-binding proteins. Their targets build peptidoglycan, making wall biology central to their activity.
Gram stain: A differential staining procedure that classifies bacteria by how their cell envelopes retain dyes. This staining method supplies the defining test for this group.
Alcohol decolorization: The Gram-staining step in which alcohol or acetone removes dye from cells that do not retain the primary stain. Thick peptidoglycan resists dye loss during this decisive step.
Clinical microbiology: The study of microorganisms that cause disease and their detection in clinical samples. Gram reaction helps describe organisms in specimens and laboratory reports.
Vancomycin: A glycopeptide antibiotic that inhibits cell-wall synthesis by binding peptidoglycan precursors. It acts on exposed wall-building targets in susceptible Gram-positive bacteria.
Crystal violet: A purple dye used as the primary stain in Gram staining and in other applications. It is the dye retained in the thick cell wall during the procedure.
Safranin: A red counterstain used in Gram staining after the decolorization step. It colors decolorized cells, while Gram-positive cells remain predominantly purple.
