Lamotrigine

Lamotrigine is a prescription medicine used to treat epilepsy and to help prevent mood episodes in bipolar disorder. It acts mainly by reducing excessive electrical signaling in the brain.

Voltage-gated sodium channel: A membrane protein that opens in response to voltage changes and enables rapid electrical signaling in excitable cells. Lamotrigine inhibits sodium-channel activity, reducing repetitive neuronal firing.
Epilepsy: A neurological disorder characterized by an enduring predisposition to epileptic seizures. Lamotrigine is used alone or with other medicines to treat several seizure types.
Lithium: A mood-stabilizing medication used to treat and prevent episodes of bipolar disorder. Lithium has broader evidence for mania and suicide prevention; lamotrigine is more useful for depressive relapse prevention.

Valproate: An antiseizure medicine also used to treat manic episodes and prevent bipolar mood episodes. Valproate interacts with lamotrigine and increases its exposure, requiring a lower, slower dosing schedule.
Glucuronidation: A metabolic process that attaches glucuronic acid to compounds, often making them easier to eliminate. Lamotrigine is primarily cleared through glucuronidation in the liver.
Pregnancy and epilepsy: The clinical management of seizures and antiseizure medicines during pregnancy. Lamotrigine concentrations can fall during pregnancy and rise again after delivery, affecting seizure control and dosing.
