Levodopa

Levodopa is an amino-acid precursor that crosses the blood–brain barrier and is converted into dopamine. It is a principal treatment for the motor symptoms of Parkinson’s disease.

Dopamine: Dopamine is a neurotransmitter and signaling molecule involved in movement, motivation, reward, and other functions. Levodopa raises brain dopamine by serving as its immediate biochemical precursor.
Parkinson's disease: Parkinson’s disease is a progressive neurological disorder involving loss of dopamine-producing neurons and characteristic motor symptoms. Levodopa is the most effective medication for the disease’s core motor symptoms.
Dopamine agonist: A dopamine agonist is a drug that activates dopamine receptors without first being converted into dopamine. Unlike levodopa, these drugs stimulate receptors directly rather than supplying dopamine’s precursor.

Levodopa-induced dyskinesia: Levodopa-induced dyskinesia is involuntary, often choreiform movement associated with long-term levodopa treatment. Repeated dopamine replacement can eventually produce troublesome involuntary movements.
Monoamine oxidase B inhibitor: A monoamine oxidase B inhibitor is a drug that slows the breakdown of dopamine in the brain. These medicines preserve existing dopamine, whereas levodopa supplies material for making more.
Oleh Hornykiewicz: Oleh Hornykiewicz was an Austrian neurologist whose research linked dopamine depletion to Parkinson’s disease. His findings provided a biochemical rationale for treating Parkinson’s with levodopa.
Blood–brain barrier: The blood–brain barrier is a selective interface that regulates passage of substances from blood into the central nervous system. Levodopa crosses this barrier, while dopamine itself crosses poorly.
Wearing-off phenomenon: The wearing-off phenomenon is the return of Parkinson’s symptoms as a medication dose loses effect before the next dose. As disease progresses, a levodopa dose may provide a shorter period of symptom relief.
