Optogenetics

Optogenetics uses genetically introduced, light-sensitive proteins to control the activity of selected cells with light. It enables precise control over cell type, timing, and location, especially in neuroscience.

Channelrhodopsin: A light-gated ion channel that opens in response to blue light, often depolarizing cells. It is the best-known actuator for exciting neurons with light.
Promoter (genetics): A DNA sequence that helps initiate transcription of a gene. Promoters can restrict optogenetic protein expression to particular cell populations.
Neural circuit: A set of interconnected neurons whose activity contributes to a function or behavior. Optogenetic perturbations test how particular circuit elements contribute to function.
Francis Crick: A British molecular biologist who co-discovered DNA's double-helical structure and proposed using light to control neurons. His 1999 proposal articulated the challenge of controlling selected neurons with light.
Electrophysiology: The study and measurement of electrical properties in cells and tissues. It measures electrical activity directly, while optogenetics can perturb it with light.
Halorhodopsin: A light-driven chloride pump that can hyperpolarize cells when illuminated. It provides an inhibitory counterpart to excitatory channelrhodopsins.
Cre-Lox recombination: A site-specific recombination system that rearranges DNA at lox sites in cells expressing Cre recombinase. It enables conditional expression of optogenetic tools in selected cell types.
Causal inference: Methods for estimating whether changes in one variable cause changes in another. Selective stimulation helps distinguish causal circuit effects from mere correlations.
Gero Miesenböck: An Austrian-born neuroscientist who developed early methods for genetically targeted optical control of neurons. His work demonstrated optical control before microbial channelrhodopsins became standard tools.
Chemogenetics: A method for controlling cell activity through engineered receptors activated by selected chemicals. It offers cell-targeted control without the precise timing of light stimulation.
