Proteinogenic amino acid

A proteinogenic amino acid is an amino acid incorporated into proteins by cellular translation. The standard set comprises 20 canonical amino acids and, in some organisms, selenocysteine and pyrrolysine.

Amino acid: An organic molecule containing amino and carboxyl functional groups; many serve as protein building blocks. Proteinogenic amino acids are a subset defined by their incorporation into proteins.
Translation (biology): The cellular process that synthesizes a protein by decoding messenger RNA on a ribosome. Proteinogenic amino acids are incorporated into chains during this process.
Protein engineering: The design or modification of proteins to alter their properties or functions. Changing proteinogenic amino acids can tune stability, activity, and binding.
Post-translational modification: A chemical change to a protein made after its ribosomal synthesis. Modified residues arise after incorporation and are not separate amino acids specified at translation.
Genetic code: The rules that map nucleotide sequences in messenger RNA to amino acids and stop signals. Codons in this code specify which proteinogenic amino acid translation adds.
Transfer RNA: An RNA molecule that carries a specific amino acid and pairs its anticodon with a messenger RNA codon. Transfer RNAs deliver amino acids whose identities match the translated codons.
Site-directed mutagenesis: A method for introducing a chosen DNA sequence change at a specific site. It enables controlled substitutions between proteinogenic amino acids in a protein.
Non-proteinogenic amino acid: An amino acid that is not incorporated into proteins by standard cellular translation. It contrasts with amino acids defined by their role as translation building blocks.
Peptide bond: A covalent amide bond linking the carboxyl group of one amino acid to the amino group of another. Translation joins proteinogenic amino acids into polypeptide chains through these bonds.
Aminoacyl-tRNA synthetase: An enzyme that attaches a specific amino acid to its corresponding transfer RNA. These enzymes establish the amino-acid identity carried into the ribosome.
