Ranitidine

Ranitidine is a histamine H₂-receptor antagonist that reduces stomach acid secretion. It was used to treat peptic ulcers, gastroesophageal reflux, and related conditions before safety concerns led to its withdrawal or suspension in many countries.

Histamine H₂ receptor: A G protein-coupled receptor that stimulates gastric acid secretion when activated by histamine. Ranitidine reduces acid by blocking this receptor on acid-secreting stomach cells.
Peptic ulcer disease: A condition in which sores form in the stomach or upper small-intestinal lining. Reducing acid helped ulcers heal and relieved ulcer-related symptoms.
Cimetidine: The first widely used histamine H₂-receptor antagonist, introduced for peptic-ulcer treatment. Like ranitidine, it blocks H₂ receptors, but cimetidine has more drug interactions and endocrine effects.
N-Nitrosodimethylamine: A potent nitrosamine contaminant classified as probably carcinogenic to humans. Testing found that ranitidine could contain or generate this contaminant under certain conditions.
James Black: A Scottish pharmacologist who helped develop propranolol and the first clinically useful histamine H₂-receptor antagonist. Black’s H₂-receptor research established the pharmacological strategy that led to ranitidine.
Parietal cell: A stomach epithelial cell that secretes hydrochloric acid and intrinsic factor. These cells receive the histamine signal that ranitidine blocks.
Gastroesophageal reflux disease: A chronic condition in which stomach contents reflux into the esophagus and cause troublesome symptoms or complications. Ranitidine was used to reduce acid exposure in reflux.
Famotidine: A histamine H₂-receptor antagonist used to reduce gastric acid secretion. Famotidine became a common alternative after ranitidine was withdrawn or suspended.
Drug recall: A regulatory or manufacturer action that removes or corrects a medicinal product because of a quality, safety, or labeling problem. Ranitidine products were recalled or withdrawn after contamination concerns emerged.
GlaxoSmithKline: A British multinational pharmaceutical company formed through the merger of Glaxo Wellcome and SmithKline Beecham. Glaxo developed and marketed ranitidine, widely sold under the brand name Zantac.
