Roger Y. Tsien

Roger Y. Tsien was an American biochemist who engineered fluorescent proteins and developed calcium-sensitive fluorescent indicators for imaging living cells. He shared the 2008 Nobel Prize in Chemistry for the discovery and development of green fluorescent protein.

Green fluorescent protein: A fluorescent protein first isolated from the jellyfish Aequorea victoria, widely used to mark cells and biological molecules. Tsien expanded its colors and uses, helping turn it into a versatile cellular marker.
Fura-2: A fluorescent dye whose excitation spectrum shifts when it binds calcium ions. Tsien developed this ratiometric indicator to measure calcium concentration in living cells.
University of Cambridge: A public research university in Cambridge, England, founded in 1209. Tsien studied chemistry there as an undergraduate and completed his doctorate.
Osamu Shimomura: A Japanese organic chemist who discovered and isolated green fluorescent protein from Aequorea victoria. Shimomura’s isolation of the protein supplied the starting material for Tsien’s engineering work.
Fluorescent protein: A protein that absorbs light at one wavelength and emits it at a longer wavelength. Tsien’s protein engineering helped make this broader class of biological markers useful in cells.
Indo-1: A calcium-binding fluorescent dye whose emission wavelength changes upon binding calcium. Its wavelength shift provided another way to quantify intracellular calcium.

Martin Chalfie: An American developmental biologist who demonstrated green fluorescent protein as a gene-expression marker in living organisms. Chalfie established a biological use that Tsien later broadened through protein engineering.
Fluorescence microscopy: Microscopy that uses fluorescent molecules to visualize structures or processes in a specimen. Engineered fluorescent proteins became central labels for imaging cellular structures and activity.
Cameleon: A genetically encoded calcium indicator combining fluorescent proteins with a calcium-sensitive binding component. Tsien’s group developed this sensor to report calcium changes through altered energy transfer.
