Safrole

Safrole is a naturally occurring phenylpropene found in sassafras and other plants. Its use in fragrance manufacture and as a chemical precursor is shaped by its carcinogenicity and legal controls.

Phenylpropene: A class of aromatic compounds with a three-carbon side chain attached to a benzene ring. Safrole belongs to this structural family, whose members often occur in plant essential oils.
Sassafras: A genus of aromatic trees native to eastern North America and eastern Asia. The roots and bark of some sassafras species are natural sources of safrole.
Perfumery: The art and industry of creating fragrances for personal and household products. Safrole’s sweet, spicy odor once made it useful in fragrance formulations.
Carcinogenicity: The capacity of an agent to cause cancer under particular conditions of exposure. Safrole causes liver tumors in experimental animals and is treated as a potential human carcinogen.
Methyl eugenol: A naturally occurring aromatic compound found in some essential oils and associated with genotoxic and carcinogenic hazards. It shares safrole’s plant-oil context and toxicological scrutiny, but has a different structure.
Benzodioxole: A fused aromatic ring system containing a methylenedioxy bridge. This ring system is the distinctive structural feature of safrole.
Sassafras albidum: An eastern North American tree historically valued for aromatic roots and bark. Its root-bark oil was a major historical source of safrole.
Heliotropin: A fragrant aromatic aldehyde also known as piperonal, used in perfumes and flavorings. It is a downstream chemical related to safrole and has its own fragrance applications.
Liver cancer: A group of cancers that originate in the liver, including hepatocellular carcinoma. Liver tumors observed in safrole-exposed animals underpin major safety concerns.
Anethole: An aromatic compound responsible for the characteristic anise-like scent of fennel and anise. It is a widely used fragrance and flavor compound without safrole’s precursor-control profile.
