Structure–activity relationship

A structure–activity relationship describes how changes in a compound’s chemical structure alter its biological effects, such as potency, selectivity, or toxicity.

Pharmacophore: The molecular features and spatial arrangement required for a compound to produce a particular biological effect. It identifies the structural features linked to activity within a structure–activity series.
Medicinal chemistry: The discipline that designs and optimizes compounds for therapeutic use. Structure–activity analysis is a central method for optimizing drug candidates.
Lead optimization: The process of improving a promising compound’s potency, selectivity, safety, and drug-like properties. Medicinal chemists use activity trends to choose which structural changes to test.
Activity cliff: A sharp difference in biological activity between structurally similar compounds. Activity cliffs expose cases where small structural changes defeat simple trend-based predictions.
Molecular docking: A computational method that predicts how a molecule may bind to a biological target. Docking offers structural hypotheses for activity differences among related compounds.
