Sulfonamide

Sulfonamides are synthetic antimicrobial drugs that inhibit bacterial folate synthesis. They include the first widely used systemic antibacterial medicines.

Folate synthesis: A biochemical pathway that produces folate and its derivatives from simpler precursors. Sulfonamides interrupt this pathway in bacteria by competing for an essential precursor.
Gerhard Domagk: A German physician and pathologist who demonstrated the antibacterial effects of Prontosil in 1932. His experiments established sulfonamide therapy as a practical treatment for bacterial infections.
Sulfamethoxazole: A sulfonamide antibiotic commonly combined with trimethoprim. This widely used member anchors combination therapy for several bacterial and opportunistic infections.
Sulfonamide resistance: Bacterial resistance to sulfonamide drugs, commonly caused by altered drug targets or increased para-aminobenzoic acid production. Resistance directly limits treatment by undermining the drugs’ competition at folate synthesis.
Dihydropteroate synthase: A bacterial enzyme that joins para-aminobenzoic acid to a pterin precursor during folate synthesis. Sulfonamides inhibit this enzyme by mimicking para-aminobenzoic acid.
Prontosil: A red azo dye that is converted in the body into the active sulfonamide sulfanilamide. This early drug revealed the antibacterial potential of the sulfonamide group.
Trimethoprim/sulfamethoxazole: A fixed-dose antibacterial combination that blocks two successive steps in microbial folate metabolism. The combination illustrates how sulfonamide activity can be strengthened by sequential pathway inhibition.
Antimicrobial resistance: The ability of microbes to survive exposure to medicines that would otherwise inhibit or kill them. Sulfonamide resistance is one specific instance of this broader clinical problem.
Para-aminobenzoic acid: An aromatic compound that bacteria use as a precursor in folate synthesis. Its structural resemblance to sulfonamides explains their competitive inhibition of folate production.
Sulfanilamide: A synthetic antibacterial drug and the active metabolite of Prontosil. Its identification as Prontosil’s active component enabled simpler sulfonamide medicines.
