Tadalafil

Tadalafil is a long-acting phosphodiesterase type 5 inhibitor used to treat erectile dysfunction, benign prostatic hyperplasia, and pulmonary arterial hypertension.

Phosphodiesterase type 5: Phosphodiesterase type 5 is an enzyme that breaks down cyclic guanosine monophosphate in smooth muscle. Tadalafil inhibits this enzyme, slowing cyclic GMP breakdown.
Erectile dysfunction: Erectile dysfunction is the persistent inability to achieve or maintain an erection sufficient for satisfactory sexual activity. Tadalafil improves erectile response to sexual stimulation by increasing penile blood flow.
Avanafil: Avanafil is a phosphodiesterase type 5 inhibitor used to treat erectile dysfunction. It is a shorter-acting alternative in the same drug class.

Benign prostatic hyperplasia: Benign prostatic hyperplasia is noncancerous enlargement of the prostate that can obstruct urinary flow. Daily tadalafil can reduce urinary symptoms associated with this condition.
Alprostadil: Alprostadil is a prostaglandin medication used to treat erectile dysfunction and certain neonatal heart conditions. It can treat erectile dysfunction through a different mechanism, including injection or urethral administration.
Half-life: A drug’s half-life is the time required for its concentration in the body to fall by half. Tadalafil’s roughly 17.5-hour half-life supports effects lasting longer than those of many other PDE5 inhibitors.
Nitric oxide signaling: Nitric oxide signaling uses the gaseous molecule nitric oxide to regulate processes including vascular smooth-muscle relaxation. Tadalafil amplifies cyclic GMP signaling initiated by nitric oxide; it does not itself trigger that pathway.
Pulmonary arterial hypertension: Pulmonary arterial hypertension is a progressive condition involving high pressure in the arteries carrying blood from the heart to the lungs. Tadalafil relaxes pulmonary vessels and can improve exercise capacity in this disease.
