Temazepam

Temazepam is a benzodiazepine medication prescribed mainly for short-term treatment of insomnia. It enhances signaling by GABA, an inhibitory neurotransmitter, through GABA-A receptors.

GABA-A receptor: A ligand-gated chloride channel activated by GABA and targeted by benzodiazepines. Temazepam binds receptor sites that strengthen GABA-driven inhibition.
Insomnia: A condition involving persistent difficulty initiating or maintaining sleep despite adequate opportunity. Temazepam is prescribed primarily to relieve short-term insomnia.
Benzodiazepine dependence: Physical or psychological reliance on benzodiazepines, sometimes accompanied by withdrawal symptoms when use stops. Repeated temazepam use can lead to dependence, making duration and discontinuation important.
Zolpidem: A nonbenzodiazepine hypnotic that acts at benzodiazepine-sensitive sites on GABA-A receptors. It treats insomnia through a related receptor site but has a different chemical structure.
Sleep architecture: The structure and progression of sleep stages across a night. Sedatives can change sleep-stage patterns as well as the ability to fall asleep.
Gamma-aminobutyric acid: The principal inhibitory neurotransmitter in the mammalian central nervous system. Temazepam amplifies the effects of this neurotransmitter rather than replacing it.
Sleep-onset insomnia: Insomnia characterized chiefly by difficulty falling asleep at the start of the night. Temazepam's sedative effect can help shorten the time needed to fall asleep.
Benzodiazepine withdrawal syndrome: A range of symptoms that can follow reduction or cessation after sustained benzodiazepine exposure. Abruptly stopping regular temazepam use can provoke withdrawal, including seizures in severe cases.
Triazolam: A short-acting benzodiazepine prescribed for short-term insomnia treatment. Its shorter action contrasts with temazepam's intermediate duration.
Circadian rhythm: An approximately 24-hour biological cycle that coordinates physiological processes with day and night. Circadian misalignment can cause sleep problems that temazepam does not correct.
