Tigecycline

Tigecycline is a glycylcycline antibiotic that binds the bacterial 30S ribosomal subunit, inhibiting protein synthesis. It is used for certain serious bacterial infections, including infections resistant to other antibiotics.

30S ribosomal subunit: The smaller subunit of bacterial ribosomes, responsible for decoding messenger RNA during protein synthesis. Tigecycline binds this subunit and blocks aminoacyl-tRNA from entering the ribosome.
Complicated skin and skin-structure infection: A severe infection involving skin or underlying tissues, often requiring systemic antibiotic treatment. Tigecycline is approved to treat certain complicated infections of this kind.
Eravacycline: A fluorocycline antibiotic related to tigecycline and used for selected bacterial infections. This later tetracycline derivative has different indications and dosing despite a related mechanism.
FDA boxed warning: A prominent United States drug-label warning reserved for serious or life-threatening risks. Tigecycline carries a boxed warning about increased all-cause mortality observed in clinical trials.
Protein synthesis inhibitor: An antimicrobial agent that stops bacteria from producing proteins by disrupting translation. Tigecycline belongs to this broad class of antibiotics.
Complicated intra-abdominal infection: An infection within the abdomen involving an abscess, perforation, or spread beyond a single organ. Tigecycline is used for selected complicated intra-abdominal infections.
Omadacycline: A modern aminomethylcycline antibiotic used for community-acquired infections including pneumonia and skin infections. Unlike tigecycline, omadacycline is available in oral formulations.
Antimicrobial resistance: The ability of microorganisms to survive exposure to drugs that once inhibited or killed them. Tigecycline was developed partly to address tetracycline resistance, but resistance can also emerge to it.
Tetracycline: A broad-spectrum antibiotic that inhibits bacterial protein synthesis by binding the 30S ribosomal subunit. Tigecycline was designed to retain tetracycline activity against common resistance mechanisms.
Community-acquired bacterial pneumonia: Pneumonia acquired outside a hospital or long-term care facility. Tigecycline has an indication for community-acquired bacterial pneumonia in adults.
