Triazolam

Triazolam is a short-acting benzodiazepine medication prescribed for short-term treatment of insomnia. It enhances GABA-mediated inhibition in the central nervous system and can cause amnesia, dependence, and withdrawal.

GABA receptor: A family of receptors activated by the inhibitory neurotransmitter gamma-aminobutyric acid. Triazolam acts at GABA type A receptors to strengthen inhibitory signaling.
Insomnia: A disorder involving persistent difficulty falling asleep, staying asleep, or obtaining restorative sleep despite adequate opportunity. Short-term insomnia treatment is triazolam's principal approved use.
Anterograde amnesia: An inability to form lasting memories of events occurring after a particular point or intervention. Triazolam can impair memory formation after a dose, especially during nighttime activity.
Temazepam: A benzodiazepine medication used for short-term treatment of insomnia, with a longer duration than triazolam. Its longer action can suit sleep maintenance better but may increase next-day effects.
GABAergic: Describes neurons, drugs, or processes that use or affect gamma-aminobutyric acid signaling. Triazolam is a GABAergic drug, reducing activity across neural circuits.
Sleep-onset insomnia: Insomnia characterized chiefly by difficulty falling asleep at the beginning of the night. Triazolam's rapid onset makes it suited to difficulty initiating sleep.
Benzodiazepine dependence: Physical adaptation to benzodiazepines that can produce withdrawal symptoms when use is reduced or stopped. Repeated triazolam use can lead to dependence despite its short duration of action.
Zolpidem: A nonbenzodiazepine hypnotic that acts at benzodiazepine-sensitive GABA type A receptor sites. It is a commonly compared prescription sleep medicine with a different chemical structure.
Positive allosteric modulator: A substance that increases a receptor's response to its natural ligand by binding at a separate site. Triazolam amplifies GABA's action rather than directly activating its receptor.
Cognitive behavioral therapy for insomnia: A structured treatment that changes sleep-related behaviors and thoughts through techniques such as stimulus control and sleep restriction. It is a preferred durable treatment, unlike triazolam's short-term symptom relief.
