Promoter

A promoter is a DNA region where transcription machinery assembles to initiate transcription. Its sequence and location help determine when, where, and how strongly a gene is transcribed.

Transcription initiation: The stage of gene expression in which RNA polymerase begins synthesizing an RNA molecule from a DNA template. Promoter-bound machinery assembles the components that begin this process.
DNA sequence: The ordered series of nucleotide bases along a DNA molecule. Promoter identity depends on the arrangement of bases in its DNA.
Promoter trapping: A genetic method that identifies active promoters by linking their activity to a detectable reporter gene. It reveals promoter activity in cells without requiring prior knowledge of the promoter sequence.
Operator (genetics): A DNA site, commonly in prokaryotes, where a regulatory protein binds to control transcription. Operators regulate transcription through repressor or activator binding rather than serving as the initiation region.
Gene expression: The production of a functional RNA or protein from the information encoded in a gene. Promoter activity is a major control point for how much RNA a gene produces.
RNA polymerase: An enzyme that synthesizes RNA by copying a DNA or RNA template. Promoters help position RNA polymerase at the start of a gene.
TATA box: A short DNA sequence motif found in some promoters that helps position transcription machinery. It is a recognizable core-promoter element in many eukaryotic genes.
Reporter gene: A gene whose measurable product is used to indicate the activity of a regulatory sequence or cellular process. Reporter genes make promoter-driven transcription experimentally visible.
Silencer (DNA): A regulatory DNA sequence that reduces gene transcription when bound by appropriate proteins. Silencers repress expression, whereas promoters provide the transcription-initiation site.
Cis-regulatory element: A noncoding DNA sequence that regulates expression of a nearby gene. Promoters are one class of cis-regulatory element.
