Ramelteon

Ramelteon is a prescription sleep medicine that activates MT1 and MT2 melatonin receptors. It is used to treat insomnia characterized by difficulty falling asleep.

Melatonin receptor: A G protein-coupled receptor that responds to melatonin and helps regulate circadian rhythms and sleep. Ramelteon works by activating the MT1 and MT2 subtypes of this receptor.
Sleep-onset insomnia: Difficulty initiating sleep despite having adequate opportunity and circumstances for sleep. This is the insomnia symptom ramelteon is specifically used to treat.

Insomnia: A sleep disorder involving persistent difficulty falling asleep, staying asleep, or obtaining restorative sleep despite adequate opportunity. Ramelteon treats a specific presentation within this broader sleep disorder.
Z-drugs: Nonbenzodiazepine hypnotics, including zolpidem, that act at benzodiazepine sites on GABA-A receptors. These common insomnia drugs use a different receptor system from ramelteon.
Agonist: A substance that binds to a receptor and activates it to produce a biological response. Ramelteon acts as an agonist at both MT1 and MT2 receptors.
