Warfarin

Warfarin is an oral anticoagulant that inhibits vitamin K recycling, reducing the production of functional vitamin K-dependent clotting factors.

Vitamin K epoxide reductase: An enzyme that recycles vitamin K epoxide into its reduced form, enabling vitamin K-dependent protein activation. Warfarin inhibits this enzyme, limiting the reduced vitamin K needed to activate clotting factors.
Atrial fibrillation: An irregular heart rhythm that can allow blood clots to form in the atria and travel to the brain. Warfarin can reduce stroke risk when atrial fibrillation raises the chance of embolic clots.
Direct oral anticoagulant: An oral anticoagulant that directly inhibits thrombin or factor Xa rather than vitamin K recycling. These drugs usually need less routine monitoring but are not suitable substitutes in every indication.
Sweet clover disease: A bleeding disorder in cattle caused by eating spoiled sweet clover containing anticoagulant compounds. The livestock outbreak revealed the biological effect that led researchers toward warfarin.
Vitamin K cycle: A biochemical cycle that regenerates reduced vitamin K for activating vitamin K-dependent proteins. The drug's target sits in this cycle, so blocking it reduces functional clotting factors.
Venous thromboembolism: A condition involving deep-vein thrombosis, pulmonary embolism, or both. Warfarin is used to treat and prevent recurrence of these venous clots.
Heparin: A family of anticoagulant drugs that enhances antithrombin activity and acts rapidly. Heparin can provide rapid anticoagulation while warfarin's effect develops gradually.
Dicoumarol: A natural anticoagulant formed from coumarin derivatives in spoiled sweet clover. Its discovery established the chemical lineage from the cattle disease to warfarin.
Coagulation factor II: Prothrombin, a vitamin K-dependent plasma protein converted into thrombin during blood coagulation. Its reduced production is central to warfarin's anticoagulant effect.
Mechanical heart valve: An artificial heart valve made from durable materials that can promote clot formation without anticoagulation. Warfarin is commonly used to prevent thrombosis on mechanical valves.
