Knowra Zolpidem Zolpidem Zolpidem is a sedative-hypnotic medication prescribed primarily for short-term treatment of insomnia. It enhances signaling at GABA-A receptors, especially receptor subtypes associated with sleep.
GABA-A receptor : A ligand-gated chloride channel activated by gamma-aminobutyric acid, the brain’s principal inhibitory neurotransmitter. Zolpidem binds a site on this receptor complex and strengthens inhibitory signaling.
Insomnia : Insomnia is persistent difficulty initiating sleep, maintaining sleep, or obtaining restorative sleep despite adequate opportunity. Zolpidem is prescribed to relieve insomnia symptoms, usually for limited periods.
Complex sleep behaviors : Complex sleep behaviors are activities such as walking, eating, or driving performed while not fully awake, often with later amnesia. Zolpidem can rarely trigger these dangerous behaviors, prompting a boxed warning.
Benzodiazepines : Benzodiazepines are drugs that enhance GABA-A receptor activity and are used for anxiety, seizures, insomnia, and other conditions. They share zolpidem’s receptor site but generally act across a broader range of receptor subtypes.
Benzodiazepine site : An allosteric binding site on certain GABA-A receptor subtypes that accepts benzodiazepines and related drugs. Zolpidem acts at this site, though its receptor-subtype preference differs from many benzodiazepines.
Immediate-release zolpidem : A zolpidem formulation designed to release the drug promptly after administration. This formulation is generally used when the main difficulty is falling asleep.
Anterograde amnesia : Anterograde amnesia is impaired formation of new memories after a drug or event takes effect. Memory gaps can accompany zolpidem use, particularly when users remain awake after taking it.
Zaleplon : Zaleplon is a short-acting nonbenzodiazepine hypnotic prescribed for difficulty falling asleep. It is another Z-drug, with a shorter duration than zolpidem.
GABA : Gamma-aminobutyric acid is the main inhibitory neurotransmitter in the adult human central nervous system. Zolpidem amplifies the inhibitory effects of GABA rather than activating its receptor by itself.
Extended-release zolpidem : A zolpidem formulation that releases drug in phases to extend exposure through the night. Its later release phase is intended to help with both sleep onset and sleep maintenance.
